<p dir="RTL" style="text-align: justify;"> سالهاست که توجه محققین بر روی یافتن ترکیبات آنتی اکسیدانی که مانع از اکسیداسیون <span dir="LTR">LDL</span> شود بدون اینکه اثرات مخربی داشته باشد معطوف گردیده است. در برخی از ویتامینها مثل ویتامین های <span dir="LTR">C</span> و<span dir="LTR">E</span>خاصیت آنتی اکسیدانی علیه اکسیداسیون <span dir="LTR">LDL</span> کاملا مشخص شده است.اسانسهای فرار از جمله فرآورده های گیاهی هستند که بدلیل خاصیت لیپوفیلیک توانایی وارد شدن بداخل ذره <span dir="LTR">LDL</span> را دارند و اثرات آنتی اکسیدانی این ترکیبات هنوز بخوبی روشن نشده است. هدف از این تحقیق بررسی امکان استفاده از فرآورده های گیاهی نظیر اسانسهای فرار به منظور پیشگیری یا درمان احتمالی آترواسکلروز میباشد.با توجه به اثر محتمل اسانسهای فرار بر روی تمایل <span dir="LTR">LDL</span> به رسپتورش میتوان این تمایل را تحت تأثیر قرار داد و از تغییرات<span dir="LTR">LDL</span> ممانعت کرد و درنتیجه باآتروژنز مقابله نمود.</p>
<p dir="RTL" style="text-align: justify;"> در این تحقیق خواص آنتی اکسیدانی غلظتهای مختلف 3 نوع اسانس فرار بنامهای : تیمول و پاراسیمول و لیمونن وهمچنین ویتامین <span dir="LTR">C</span> بر روی میزان تمایل <span dir="LTR">LDL</span> طبیعی و اکسیده (با یونهای <span dir="LTR">Cu+2</span>) به رسپتور مربوطه در سلولهای بافت آدرنال گوسفند در حضور <span dir="LTR">LDL</span> نشاندار شده با یک ماده فلوئورسنت بنام فلوئورسین ایزو تیوسیانات<span dir="LTR">(FITC)</span> مورد بررسی قرار گرفته است. برای این منظور پس از جداسازی محلول <span dir="LTR">LDL</span> از سرم نرمال و انکوباسیون آن با ترکیبات فوق <span dir="LTR">,</span> واکنش اکسیداسیون بر روی آن انجام گرفت و مقداری محلول <span dir="LTR">LDL</span> با <span dir="LTR">FITC</span> نشاندار گردید و سپس همه محلولهای فوق به محیط شامل سلولهای بافت آدرنال (که حاوی رسپتور <span dir="LTR">LDL</span> هستند) اضافه شد و شدت فلورسانس محلول رویی مخلوط آزمایش پس از سانتریفوژ در اسپکتروفلوریمتر خوانده شد. همچنین اثرات تؤام بهترین غلظت آنتی اکسیدانی ویتامین <span dir="LTR">C</span> ( بدست آمده در این پروژه) با هر یک از غلظتهای مختلف سه اسانس فوق نیز ارزیابی گردیده است.</p>
<p dir="RTL" style="text-align: justify;"> نتایج بدست آمده نشان میدهد که ویتامین <span dir="LTR">C</span> در غلظت 100 میکرومولار دارای بیشترین اثر برروی افزایش تمایل <span dir="LTR">LDL</span> به رسپتورش میباشد( برای <span dir="LTR">LDL</span> طبیعی 5/18 درصد و برای <span dir="LTR">LDL</span> اکسیده 62 درصد) واز میان اسانسهای مورد آزمایش تیمول دارای بیشترین اثر است( برای <span dir="LTR">LDL</span> طبیعی 27 درصد و برای <span dir="LTR">LDL</span> اکسیده 50 درصد). همچنین در بررسی اثرات همزمان هر یک از این اسانسها با مؤثرترین غلظت ویتامین <span dir="LTR">C</span> (100 میکرومولار) باز هم تیمول بالاترین اثر مثبت رابر روی تمایل <span dir="LTR">LDL</span> به رسپتورش داشته است.</p>
<p dir="RTL" style="text-align: justify;"> این یافته ها بیانگر اینست که احتمالا" ترکیبات فوق با اثر برروی اتصال <span dir="LTR">LDL</span> به رسپتور مربوطه میتوانند از ایجاد آترواسکلروز جلوگیری کرده و بعنوان آنتی اکسیدان مورد توجه قرار گیرند.</p>

<p style="text-align: justify;"> For several years , scientists tried to find antioxidant compounds that<br>
inhibit the low density lipoprotein(LDL) oxidation reaction but have no<br>
destructive effects. Vitamin C and vitamin E can inhibit LDL oxidation.<br>
Volatile oils posses herbal sources and because of lipophilic properties can<br>
enter LDL and their antioxidant effects have not been known. The aim of<br>
this study is possible use of herbal products such as volatile oils in preventing<br>
or treating atherosclerosis. Because the probable effects of volatile oils on<br>
the affinity of LDL to its receptor , this affinity can be influenced and the <br>
modification of LDL and atherogenesis be prevented.</p>
<p style="text-align: justify;"> In this study , antioxidant properties of various concentrations of three<br>
volatile oils include:P-cymol, Limonene and Thymol , and vitamin C on the<br>
affinity of natural-LDL and oxidized-LDL(with Cu+2) to its receptor in bovine<br>
adrenal tissue cells have been investigated in the presence of labeled-LDL<br>
with fluoresein isothiocyanate (FITC). First, after purification of LDL-solvent of<br>
normal serum and incubation it with these compounds, oxidation reaction<br>
performed and LDL-solvent was labeled with FITC, and then all solvents were<br>
added to the medium that contains adrenal tissue cells(with LDL-receptor).<br>
Finally, fluorescence intensity of supernatant of each sample was<br>
determined in spectrofluorometer. Also, the effects of the best<br>
concentration of vitamin C (obtained in this study) with each one of these<br>
compounds was studied.</p>
<p style="text-align: justify;"> Results showed that vitamin C in 100 micromolar concentration has the<br>
greatest effect on the increasing affinity of LDL to its receptor (for natural-<br>
LDL 18.5% and for oxidized-LDL 62%). Among the volatile oils used, “Thymol”<br>
had the best effect (for natural-LDL 27% and for oxidized-LDL 50%). In<br>
studying the effects of these three volatile oils with the best concentration of<br>
vitamin C(100 micromolar) , “Thymol” had the highest effect on this affinity.</p>
<p style="text-align: justify;"> These finding showed that some compounds probably can change the<br>
affinity of LDL to its receptor and thus may prevent formation of<br>
atherosclerosis .</p>
