fa تعیین نیمه کمی میزان بیان mRNA اینترفرون گاما با استفاده از روش RT-PCR In Vitro Semi-Quantitative Determination of Human Gamma-Interferon-mRNA Level by RT-PCR سایر تخصص هاي باليني Other Clinical Specialties پژوهشي Original <span id="ctl00_ContentPlaceHolder1_DataList1_ctl00_abstractLabel" style="font: 12px/15pt tahoma; color: rgb(45, 135, 179); text-transform: none; text-indent: 0px; letter-spacing: normal; word-spacing: 0px; white-space: normal; widows: 1; font-size-adjust: none; font-stretch: normal; background-color: rgb(247, 246, 243); -webkit-text-stroke-width: 0px;">سایتوکاینها واسطه های شیمیائی هستند که از سلولهای سیستم ایمنی ترشح شده، و بر سلولهای دیگر یا خود سلول ترشح کننده اثر می کنند. بیان متفاوت ژن سایتوکاینها در بدن مشخص کننده نوع پاسخ ایمنی (از نوع Th1 یا Th2) است که خود این پاسخ متفاوت می تواند نقش مهمی در پاتوژنز بیماریها داشته باشد. از عمده ترین سایتوکاینهای سلولهای Th1 می توان به IFN-γ و IL-2 اشاره کرد. که در پاسخهای ایمنی علیه پاتوژنهای داخل سلولی شرکت می کنند. با توجه به اینکه بیان ژن سایتوکاینها منجمله اینترفرون موقتی بوده و در یک مدت زمان محدود می باشد ، به این علت لازم است بیان ژن آنها (مقدار mRNA) توسط روشهای حساس و دقیقتری همانند RT-PCRاندازه گیری شود .بعلاوه تعیین زمان مورد نیاز جهت ماگزیمم بیان ژن می تواند در مطالعات کلونینگ و استعداد ابتلا به بیماریها در بعضی ازافراد کاربرد داشته باشد. <span id="ctl00_ContentPlaceHolder1_DataList1_ctl00_abstractLabel" style="font: 12px/15pt tahoma; color: rgb(45, 135, 179); text-transform: none; text-indent: 0px; letter-spacing: normal; word-spacing: 0px; white-space: normal; widows: 1; font-size-adjust: none; font-stretch: normal; background-color: rgb(247, 246, 243); -webkit-text-stroke-width: 0px;">          بعد از کشت لنفوسیتها در حضور PHA (Phytohaemagglutinin) ( با غلظت 1μg/106cell ) در زمانهای متفاوت (72،0،4،8،12،24،48 ساعت) و استخراج RNA توتال از آنها و سنتز cDNA تک رشته ای اقدام به انجام RT-PCR شد. چون در تمام ساعات انکوبه جواب مثبت بود لذا در مرحله بعدی اقدام به تیتراسیون cDNAحاصل با ضریب رقت 1.2 (رقتهای متوالی 1.2، 1.4، 1.8،و….) شد. بر روی ژل الکترفورز (2%) قطعه مورد نظر (273bp ) مشاهده شد. <span id="ctl00_ContentPlaceHolder1_DataList1_ctl00_abstractLabel" style="font: 12px/15pt tahoma; color: rgb(45, 135, 179); text-transform: none; text-indent: 0px; letter-spacing: normal; word-spacing: 0px; white-space: normal; widows: 1; font-size-adjust: none; font-stretch: normal; background-color: rgb(247, 246, 243); -webkit-text-stroke-width: 0px;">          نتایج بدست آمده از RT-PCR و Semi-quantitative-RT-PCR نشان می دهد بعد از چهار ساعت انکوباسیون لنفوسیتها با PHA ، بیشترین بیان ژن را دارند (تیتر cDNA=512) و معمولاً بعد از گذشت 24 ساعت مقدار آن به سطح اولیه (قبل از تحریک) بر می گردد. <span id="ctl00_ContentPlaceHolder1_DataList1_ctl00_abstractLabel" style="font: 12px/15pt tahoma; color: rgb(45, 135, 179); text-transform: none; text-indent: 0px; letter-spacing: normal; word-spacing: 0px; white-space: normal; widows: 1; font-size-adjust: none; font-stretch: normal; background-color: rgb(247, 246, 243); -webkit-text-stroke-width: 0px;">          با توجه به نتایج فوق و مقایسه آن با نتایج محققین دیگر می توان گفت روش RT-PCR  حساسترین روش سنجش بیان ژن جهت سایتوکاینهاست و بیان ژن اینترفرون بعد از گذشت چهار ساعت انکوبه به ماکزیمم می رسد. بنابر این مدت زمان انکوباسیون با PHA جهت بررسی اثر عوامل مختلف بر تولید و بیان ژن اینتر فرون گاما زمان ایده آلی است. <p style="font: 16px/normal "Times New Roman"; text-align: justify; color: rgb(45, 135, 179); text-transform: none; text-indent: 0px; letter-spacing: normal; word-spacing: 0px; white-space: normal; widows: 1; font-size-adjust: none; font-stretch: normal; background-color: rgb(247, 246, 243); -webkit-text-stroke-width: 0px;">Cells communicate with one another not only by cell-cell contact, but also via the elaboration of soluble mediators or cytokines. The functionally distinct repertoire of secreted cytokines of Th1/Th2 cells has been shown to play an important role in the pathogenesis of inflammatory diseases. Effector Th1 cells secretes predominantly IFN- γ and IL-2 and regulates cell-mediated immunity against intracellular pathogens. Regulatory cytokines    are likely to be secreted in small quantities in response to specific stimuli and taken by responder cells. As a result, many scientists determine cytokine’s m-RNA expression by using a more sensitive technique, RT-PCR. <p style="font: 16px/normal "Times New Roman"; text-align: justify; color: rgb(45, 135, 179); text-transform: none; text-indent: 0px; letter-spacing: normal; word-spacing: 0px; white-space: normal; widows: 1; font-size-adjust: none; font-stretch: normal; background-color: rgb(247, 246, 243); -webkit-text-stroke-width: 0px;">          The expression of IFN- γ -mRNA was studied using semiquantitative RT- PCR. Lymphocytes were stimulated by phytohaemagglutinin (PHA) (1μg/106) in culture medium (incubation time: 0,4,8,12,24,48 and72 hour) and total RNA extracted and cDNA synthesized. IFN- γ was detected by the    RT-PCR (reverse transcription-polymerase chain reaction) and semi-quantitative-RT-PCR methods, in which sequences (273bp) between two oligonucleotide primers (chosen from two exons of the IFN- γgene sequences) were amplified using a heat-stable DNA polymerase. In semi-quantitative RT-PCR we used a serial dilution (1/2,1/4…) of cDNA in order to determine the titer of cDNA for that will give visible band in 2% agarose gel  electrophoresis. <p style="font: 16px/normal "Times New Roman"; text-align: justify; color: rgb(45, 135, 179); text-transform: none; text-indent: 0px; letter-spacing: normal; word-spacing: 0px; white-space: normal; widows: 1; font-size-adjust: none; font-stretch: normal; background-color: rgb(247, 246, 243); -webkit-text-stroke-width: 0px;">          Results showed that after 4h incubation express highest level of IFN- γ- mRNA and it is stable until 24 hour after that.
