Background and Objective: Chronic Hepatitis B virus (HBV) infection is a major global health concern and a leading cause of hepatocellular carcinoma (HCC). Loss or suppression of PTEN expression contributes to the development and progression of HCC, particularly in the context of chronic HBV infection. The present study aimed to evaluate PTEN gene expression in patients with chronic HBV infection compared to healthy individuals.
Materials and Methods: The present case-control study included 50 patients with chronic HBV infection and 50 healthy controls. Peripheral blood samples were collected, and total RNA was extracted and converted to cDNA. Gene expression of PTEN relative to GAPDH was assessed by SYBR Green-based Real-Time PCR using the 2-ΔΔCt method. Statistical analysis was performed using the SPSS (version 26) software, with a significance set at P<0.05.
Results: The PTEN gene expression was significantly down-regulated in the HBV-infected group compared to controls (2-ΔΔCt = 0.074, P<0.0001). The ΔCt value of PTEN was significantly higher in the patient group, indicating reduced transcript levels. An age-related decline in PTEN expression was also observed among HBV patients.
Conclusion: The results highlight the importance of PTEN as a potential biomarker in the progression of the disease to liver cancer. Therefore, further research with a larger sample size and investigation of the molecular mechanisms involved in regulating PTEN expression seems necessary.
